邓红雨研究组长期的研究方向是病毒复制及组装释放的分子机理、病毒与宿主天然免疫系统之间的相互作用以及新型疫苗的研发。目前的主要研究对象为囊膜病毒,包括疱疹病毒和冠状病毒。截至目前已在Science Advances,Molecular Cell,PNAS,Journal of Virology,Nature Communications,Journal of Cell Biology等国际著名期刊发表论文60余篇。
研究表明约15-20%的人类癌症在病因学上与病毒感染有关。邓红雨研究组长期的研究重点是疱疹病毒γ亚家族的成员,包括Epstein-Barr病毒(EBV)和Kaposi's肉瘤相关疱疹病毒(KSHV),这两种病毒与肿瘤的发生发展密切相关,可引起鼻咽癌和恶性淋巴瘤等。以往的研究大都关注于病毒潜伏期表达的致瘤基因,而近期临床和基础研究结果都表明,裂解期复制对病毒致瘤亦至关重要。由于EBV和KSHV难以在体外进行高滴度培养并且缺乏理想的小动物模型,对其裂解期复制的研究受到了限制。邓红雨研究组同时采用了在分子水平上与EBV以及KSHV高度同源的鼠疱疹病毒68(MHV-68)为实验模型,研究致瘤疱疹病毒的组装释放以及病毒与宿主相互作用的机理。通过长期积累,已建立了一套较完整的体系,在分子、细胞和动物水平上深入系统地研究致瘤性疱疹病毒复制的关键步骤、调控机理及与宿主的相互作用。
新冠疫情爆发后,邓红雨研究组快速开展了针对冠状病毒的研究。利用在疱疹病毒研究过程中建立的针对囊膜病毒组装释放的实验体系以及病毒与宿主细胞相互作用的研究方法和经验,研究冠状病毒复制及组装释放的分子机理;同时以同属于β冠状病毒的人类冠状病毒HCoV-OC43及鼠肝炎病毒MHV-A59为模型,研究冠状病毒与宿主细胞之间的相互作用机理,为抗病毒药物的研发与设计提供新靶点和新思路。
